HDAC inhibitors upregulate PLD2 expression in breast cancer cells. a The indicated cancer cells were treated with the HDAC inhibitors TSA (400Β nM), SAHA (2Β ΞΌM), and apicidin (5Β ΞΌM) for 24Β h. The lysates were then analyzed by q-PCR and western blot using the antibody to PLD2. b MDA-MB 231 cells were cultured and labeled with [3H] myristate for 12Β h and treated with HDAC inhibitors for 1Β h after which PLD activity was measured. c MDA-MB 231 cells were treated with the indicated concentrations of SAHA for 24Β h or with 2Β ΞΌM of SAHA for the indicated time, after which PLD2 expression and acetylated histone H4 levels were assessed by western blotting. d The cells were transfected with the pGL4-PLD2 promoter and treated with the indicated HDAC inhibitors for 24Β h. The level of luciferase activity was then determined. The intensity of the indicated bands was normalized to the intensity of their respective Ξ±-tubulin bands and quantified against each other.
