BJ1108 negatively regulates CD4+ T cells differentiation without antigen specificity. Naïve CD4+ T cells and antigen presenting cells isolated from spleen and LNs of OT-II mice. Cells were cultured under Th1- and Th17-polarizing conditions with OVA323–339 (0.1 μM) in the presence or absence of a a single concentrations (10 μM) or b multiple concentrations of BJ-1108. Cells were then re-stimulated with phorbol 12-myristate 13-acetate, ionomycin, and GolgiStop for 4 h, followed by intracellular cytokine staining and flow cytometry. Representative data (mean ± SEM) of three independent experiments are shown. *p < 0.01 and ***p < 0.0001 versus vehicle
